本研究从拟南芥(Arabidopsis thaliana)gDNA中克隆了AtUGAE4基因的家族保守序列片段,反向连接在AtHSP70热激启动子和NOS终止子之间,构成含有该基因反义表达盒的表达载体pV-70brUGAE4,并经根癌农杆菌(Agrobacterium tumefaciens)介导,成功导入烟草基因组。这为探讨抑制UGAE家族基因表达对植物果胶生物合成和对离体培养细胞的细胞粘连性的影响奠定了基础。
In this study, a family of conserved sequences of AtUGAE4 gene was cloned from gDNA of Arabidopsis thaliana and ligated reversely between AtHSP70 heat shock promoter and NOS terminator to construct an expression vector pV containing the gene antisense expression cassette -70brUGAE4 and successfully introduced into tobacco genome mediated by Agrobacterium tumefaciens. This laid the foundation for exploring the effect of inhibition of UGAE family gene expression on plant pectin biosynthesis and cell adhesion to cultured cells in vitro.
