目的研究由AFP启动子控制的腺病毒载体在肝癌细胞中定向表达报告基因活性。方法采用分子生物学技术手段,将AFP启动子和绿色荧光蛋白(GFP)报告基因插入腺病毒载体质粒中,并在293细胞中重组出携带GFP基因的腺病毒AdAFP-GFP。荧光显微镜下观察GFP基因在肝癌细胞中的特异性表达。结果AdAFP-GFP能够介导GFP基因在肝癌细胞中特异性表达,多数细胞呈现荧光反应,而在正常细胞中未发现GFP的表达。结论采用AFP启动子可以实现目的基因在肝癌细胞中定向而特异性表达,这项研究应用于抗肿瘤基因治疗,将有利于显著提高治疗效果。
Objective To study the expression of reporter gene in hepatoma cells by adenovirus vector controlled by AFP promoter. Methods Using molecular biology techniques, AFP promoter and green fluorescent protein (GFP) reporter gene were inserted into adenoviral vector plasmid, and adenovirus AdAFP-GFP carrying GFP gene was recombined in 293 cells. The specific expression of GFP gene in hepatoma cells was observed under fluorescence microscope. Results AdAFP-GFP was able to mediate the specific expression of GFP gene in hepatocellular carcinoma cells. The majority of cells exhibited fluorescence response, but no GFP expression was found in normal cells. Conclusion The target gene can be specifically and specifically expressed in hepatocellular carcinoma cells by using the AFP promoter. This study will be beneficial for the treatment of anti-tumor genes and will significantly improve the therapeutic effect.
