目的:观察唐古特大黄多糖(TMP)对小鼠急性肝损伤的保护作用及探讨TMP对肝损伤的保护机理.方法:小鼠60只,随机分6组,生理盐水正常对照组,药物模型组,生理盐水模型对照组和3个给药组,每组按要求分别用CCl4156mg/kg,D-Gal-N800mg/kg,TAA550mg/kg腹腔注射(ip),造成小鼠急性肝损伤,以TMP100mg/kg,200mg/kg,400mg/kg×5d灌胃,用硫代巴比妥酸比色法测定小鼠肝组织MDA含量,用比色法测定小鼠血清的ALT,AST含量以观察TMP对小鼠急性肝损伤的保护作用.结果:TMP灌胃保护,肝组织MDA,血清ALT,AST含量明显低于对照组(P<0.05),且呈剂量依赖性.结论:TMP对CCl4,D-Gal-N,TAA引起的小鼠急性肝损伤有明显的保护作用.
Objective: To observe the protective effect of Tangut Rhubarb Polysaccharide (TMP) on acute liver injury in mice and to explore the protective mechanism of TMP on liver injury. Methods: Sixty mice were randomly divided into six groups: normal saline control group, drug model group, saline model control group and three administration groups. Each group was treated with CCl4156mg / kg, D-Gal-N800mg / kg and TAA550mg / kg intraperitoneally (ip) to induce acute hepatic injury in mice. TMP100mg / kg, 200mg / kg, 400mg / kg × 5d were given intragastrically. The liver tissue of mice was measured by thiobarbituric acid colorimetry MDA content, the contents of ALT and AST in mouse serum were determined by colorimetry to observe the protective effect of TMP on acute liver injury in mice. Results: The content of MDA, serum ALT and AST in TMP group was significantly lower than that in control group (P <0.05), and it was dose-dependent. Conclusion: TMP has obvious protective effect on acute liver injury induced by CCl4, D-Gal-N and TAA in mice.
