目的探讨雌、孕激素对淋球菌(Ng)诱导的人宫颈癌细胞胸腺基质淋巴细胞生成素(TSLP)和转化生长因子β(TGF-β)表达的影响。方法体外培养宫颈癌细胞株Hela,将获得的Hela细胞分为如下5组:对照组、Ng组、Ng+雌激素组、Ng+孕激素组、Ng+雌激素+孕激素组。分别培养24h后应用实时荧光定量PCR检测各组细胞TSLP和TGF-βmRNA表达变化情况,Western blot检测各组细胞TSLP和TGF-β蛋白水平。结果 Hela细胞在淋球菌的刺激下,细胞TSLP和TGF-βmRNA及蛋白表达水平增高(P﹤0.01);雌、孕激素单独或联合作用24h后都可以显著提高Hela细胞中TSLP和TGF-βmRNA表达水平(P﹤0.01)。结论 Hela细胞在淋球菌作用下合成TSLP和TGF-β增加;提示雌、孕可促进淋球菌诱导的TSLP和TGF-β合成。
Objective To investigate the effects of estrogen and progesterone on the expression of TSLP and TGF-β in human cervical cancer cells induced by Neisseria gonorrhoeae (Ng). Methods Hela cells were cultured in vitro. Hela cells were divided into five groups as follows: control group, Ng group, Ng + estrogen group, Ng + progesterone group, Ng + estrogen + progesterone group. After cultured for 24 hours, the expression of TSLP and TGF-βmRNA in each group were detected by real-time fluorescence quantitative PCR. The protein levels of TSLP and TGF-β in each group were detected by Western blot. Results The mRNA and protein expressions of TSLP and TGF-β in Hela cells increased significantly (P <0.01), while the expression of TSLP and TGF-βmRNA in Hela cells could be significantly increased after stimulated with Neisseria gonorrhoeae for 24 h Level (P <0.01). Conclusion Hela cells synthesize TSLP and TGF-β under the action of Neisseria gonorrhoeae. It is suggested that female and pregnant can promote Neisseria gonorrhoeae induced TSLP and TGF-β synthesis.
