建立了一种基于恒温隔绝PCR(Insulated isothermal PCR,ii PCR)技术现场检测牛羊包虫病的方法.结果显示,该方法只能检出细粒棘球绦虫,对多房棘球绦虫、肝片吸虫、细颈囊尾蚴、前后盘吸虫、日本血吸虫、金黄色葡萄球菌、伪结核杆菌无关病原不检出;检测下限为100个拷贝,重复性好.对55份牦牛源包囊样本和70份羊源包囊样本的检出率分别为61.8和4.3%,与文献报道的检测方法的符合率分别为100%;本研究建立的ii PCR方法特异性好、灵敏度高、重复性好,从核酸提取到报告检测结果仅需1小时,操作方便,可现场使用,为牛羊包虫病的现场快速诊断提供有力的工具.
A method for on-site detection of bovine and sheep echinococcosis based on Insulin isothermal PCR (PCR) was established.The results showed that this method could detect only Echinococcus granulosus, Cysticerci, Cysticercus cellulosae, Paragonimus before and after, Schistosoma japonicum, Staphylococcus aureus, Pseudoperrobacteum were not detected; the detection limit was 100 copies with good reproducibility.Among 55 yak samples and 70 The detection rates of cysts in sheep samples were 61.8 and 4.3%, respectively. The coincidence rates with the detection methods reported in the literature were 100%, respectively. The ii PCR method established in this study had good specificity, high sensitivity and good repeatability. The results of nucleic acid extraction to the report test only 1 hour, easy to operate, field use, for the rapid diagnosis of hydatid disease on-site provide a powerful tool.
