[目的]观察摄入不同剂量硫辛酸对D-半乳糖致过氧化损伤小鼠海马细胞DNA的影响。[方法]清洁级雌性昆明种小鼠按体重随机分为5组,除空白对照组外,其余4组用D-半乳糖颈背部皮下注射,1次/d,连续6周。其中1组为模型组,余3组为硫辛酸干预组;其后经口给予不同浓度硫辛酸,空白组、模型组给予同体积溶剂,同时,模型组和干预组继续给予相同剂量的D-半乳糖颈背部皮下注射,连续8周。干预结束后,用单细胞凝胶电泳法检测和评价各组小鼠海马细胞DNA损伤。[结果]模型组小鼠海马彗星细胞尾长、尾矩、Olive尾矩值均最大,对照组均最小(P<0.05),干预组各指标均小于模型组(P<0.05)。[结论]硫辛酸是一种有效的抗氧化剂,对小鼠海马细胞DNA损伤有修复作用。
[Objective] To observe the effects of different doses of lipoic acid on the DNA of hippocampal cells induced by D-galactose in mice. [Method] Clean-grade female Kunming mice were randomly divided into 5 groups according to body weight. Except the blank control group, the other 4 groups were subcutaneously injected with D-galactose on the dorsal neck once a day for 6 weeks. One group was model group and the other three groups were treated with lipoic acid intervention group. After oral administration of lipoic acid of different concentrations, the blank group and the model group were given the same volume of solvent, while the model group and the intervention group were given the same dose of D- Galactose neck subcutaneous injection, for 8 weeks. After intervention, DNA damage of hippocampal cells in each group was detected and evaluated by single cell gel electrophoresis. [Result] The tail length, tail moment and Olive tail moment of hippocampal comet cells in model group were the largest, the smallest in control group (P <0.05). The indexes in intervention group were all less than those in model group (P <0.05). [Conclusion] Lipoic acid is an effective antioxidant that can repair DNA damage in hippocampal cells of mice.
