目的探讨抗肿瘤坏死因子-α单克隆抗体(Anti-TNF-αmAb)对局灶脑缺血的作用。方法采用大白鼠局灶脑缺血尼龙线栓塞模型。将动物随机分为持续缺血24小时组和缺血6小时再灌注两大组,每组又分别应用AntiTNFαmAb与生理盐水作对照,观察梗死体积大小、梗死灶内微血管内白细胞聚集和附壁现象。结果AntiTNFαmAb能显著减少短暂脑缺血再灌注梗死体积和微血管内白细胞聚集和粘附,而对持续脑缺血动物无效。结论AntiTNFαmAb能减轻短暂脑缺血再灌注损害。
Objective To investigate the effect of anti-TNF-α mAb on focal cerebral ischemia. Methods Nylon thread embolization model of focal cerebral ischemia in rats was adopted. The animals were randomly divided into 24 hours of continuous ischemia and 6 hours of ischemia and reperfusion two groups, each group were also used anti TNF α mAb and saline as control, observe the size of the infarct volume, infarct within the microvascular endothelial cell aggregation And the phenomenon of attachment. Results Anti TNF α mAb can significantly reduce transient ischemic reperfusion infarction volume and microvascular endothelial cell aggregation and adhesion, and persistent ischemic animals invalid. Conclusion AntiTNFαmAb can reduce transient ischemic reperfusion injury.
