目的:优选四君子汤总多糖的微波提取适宜条件。方法:利用微波法进行工艺条件的改进,通过正交试验设计对总多糖提取参数进行了优选,对提取的总多糖采用红外吸收光谱和紫外吸收光谱进行分析表征。结果:微波法适合于提取四君子汤的总多糖,是一种优于传统水煎煮法、提取效率高、用时少的新的提取法。适宜的提取工艺条件是390 W的功率下,以1∶20的液固比提取6 min,提取2次,此条件下多糖含量为14.82%,比实验条件下的传统法提高了10.26%。红外光谱分析显示微波法提取的多糖的结构与传统法基本一致,且证明多糖主要来源于党参的提取物。结论:微波法提取时间短、效率高,适合于提取四君子汤的总多糖。
Objective: To optimize the conditions of microwave extraction of total polysaccharide of Sijunzi Decoction. Methods: The microwave process was used to improve the process conditions. The parameters of total polysaccharide extraction were optimized by orthogonal design. The extracted total polysaccharides were characterized by infrared absorption spectroscopy and ultraviolet absorption spectroscopy. Results: The microwave method was suitable for the extraction of total polysaccharides from Sijunzi Decoction. It is a new extraction method that is superior to traditional decoction method with high extraction efficiency and less time consuming. The optimum conditions of extraction were extraction at liquid-solid ratio of 1: 20 for 6 min and extraction time of 390 W at 2 h. Under these conditions, the content of polysaccharide was 14.82%, which was 10.26% higher than the traditional method under the experimental conditions. Infrared spectrum analysis showed that the structure of polysaccharides extracted by microwave method was basically the same as the traditional method, and the polysaccharide was mainly derived from the extract of Codonopsis pilosula. Conclusion: Microwave extraction time is short, high efficiency, suitable for extracting total polysaccharides from Sijunzi Decoction.
