目的探讨PEG修饰技术降低葛根素诱导红细胞溶血的体内研究。方法采用葛根素注射液和PEG化葛根素给予家兔每天等剂量静脉注射,连续7 d,每天取血3 mL,进行血细胞计数分析;7 d实验结束后再取血,检测红细胞渗透脆性和红细胞膜流动性。结果血细胞计数分析结果表明,PEG化葛根素组家兔的血细胞计数与空白对照组没有显著性差异,而葛根素注射液组家兔出现了血管内溶血现象;红细胞渗透脆性和红细胞膜流动性结果表明,PEG化葛根素可以降低红细胞渗透脆性和增强红细胞膜流动性。结论 PEG修饰技术可以减轻葛根素诱导血管内溶血的不良反应,更安全。
OBJECTIVE: To study the in vivo study of PEG modification on reducing hemolysis of erythrocytes induced by puerarin. Methods The rabbits were injected intravenously with puerarin and PEG-puerarin at the same dosage day by day. The blood samples were collected 3 mL daily for 7 days, then the blood count was analyzed. At the end of 7 d, blood samples were taken for detection of erythrocyte infiltration fragility and erythrocytes Membrane fluidity. Results The results of blood count analysis showed that there was no significant difference between the control group and the rabbits in the PEGylated puerarin group, whereas the rabbits in the Puerarin injection group had intravascular hemolysis. The results of erythrocyte osmotic fragility and erythrocyte membrane fluidity It is indicated that PEGylated puerarin can reduce the infiltration fragility of erythrocytes and enhance the fluidity of erythrocyte membrane. Conclusion PEG modified technique can reduce the side effects of puerarin-induced intravascular hemolysis and is safer.
