目的:研究膜联蛋白A2(ANXA2)对人结肠癌细胞SW620侵袭迁移的影响。方法:采用siRNA技术沉默ANXA2在SW620细胞中的表达,实验分为干扰组、对照组和野生组,RT-PCR法检测各组ANXA2 mRNA水平,Western blot法检测各组ANXA2、纤维蛋白溶酶(PL)、基质金属蛋白酶-1(MMP-1)、血管内皮生长因子(VEGF)蛋白的表达,Transwell侵袭实验检测各组SW620细胞穿膜能力。结果:干扰组与对照组、野生组相比,ANXA2 mRNA水平、ANXA2、PL、MMP-1、VEGF蛋白表达及SW620穿膜细胞数差异均有统计学意义(P<0.05),而对照组和野生组相比上述各指标均无统计学差异。ANXA2蛋白和PL、MMP-1、VEGF蛋白水平、穿膜细胞数呈正相关(P<0.05)。结论:沉默ANXA2基因后可能通过下调PL、MMP-1及VEGF蛋白的表达,抑制SW620细胞侵袭迁移能力起到抗肿瘤作用。
Objective: To investigate the effect of annexin A2 (ANXA2) on invasion and migration of human colon cancer cell line SW620. Methods: The expression of ANXA2 in SW620 cells was silenced by siRNA. The expression of ANXA2 mRNA in SW620 cells was detected by RT-PCR. The expression of ANXA2 and fibrinolysin PL), matrix metalloproteinase-1 (MMP-1) and vascular endothelial growth factor (VEGF) protein expression. Transwell invasion assay was used to detect the transmembrane capacity of SW620 cells in each group. Results: Compared with control group and wild group, ANXA2 mRNA expression, ANXA2, PL, MMP-1, VEGF protein expression and SW620 transmembrane cell number in the interference group were significantly different (P <0.05), while the control group and Wild group compared to the above indicators were no significant difference. ANXA2 protein and PL, MMP-1, VEGF protein levels, the number of transmembrane cells was positively correlated (P <0.05). Conclusion: The silencing of ANXA2 gene may play an anti-tumor role by down-regulating the expression of PL, MMP-1 and VEGF proteins and inhibiting the invasion and migration of SW620 cells.
