目的 研究血管内皮细胞生长因子 (vascular endothelial growth factor,VEGF)反义寡聚脱氧核苷酸在缺血性视网膜病变中的作用。 方法 以高浓度氧诱导 C5 7BL/ 6 J小鼠建立视网膜缺血性病变动物模型 ,应用血管内皮细胞生长因子反义寡聚脱氧核苷酸不同浓度进行球后注射 ,计数并比较正常和实验条件下以及不同浓度药物注射后视网膜新生血管芽细胞核数。 结果 高浓度氧诱导的小鼠视网膜发生较多的新生血管芽 ,而球后注射 VEGF反义寡聚脱氧核苷酸后视网膜新生血管芽的细胞核数明显减少 ,特别是大剂量注射后减少达 5 9.3% (P<0 .0 1)。 结论 VEGF反义寡聚脱氧核苷酸能有效抑制缺血性视网膜病变新生血管的增生
Objective To investigate the role of vascular endothelial growth factor (VEGF) antisense oligodeoxynucleotides in ischemic retinopathy. Methods The animal model of retinal ischemic lesions was established by inducing high concentration of oxygen in C5 7BL / 6 J mice. The different concentrations of vascular endothelial growth factor antisense oligodeoxynucleotides were injected into the posterior chamber to count and compare normal and experimental conditions And the number of retinal neovascular bud nuclei after injection of different concentrations of drugs. Results High concentration of oxygen induced retinal neovascularization in mice and the number of nuclei of retinal neovascularization was significantly decreased after injection of VEGF antisense oligodeoxynucleotide. Especially after high dose injection, 9.3% (P <0. 01). Conclusion VEGF antisense oligodeoxynucleotide can effectively inhibit neovascularization of ischemic retinopathy
