目的:揭示中药清热利湿饮对人永生化角质形成细胞HaCaT细胞表达IL-6mRNA、IL-8mRNA、IL-10mRNA和分泌IL-8、IL-10的作用机制。方法:以HaCaT细胞为研究对象,分别采用RT-PCR技术及ELISA方法观察中药清热利湿饮提取液对经TNF-α诱导活化的HaCaT细胞表达IL-6mRNA、IL-8mRNA、IL-10mRNA和分泌IL-8、IL-10的影响。结果:清热利湿饮对经TNF-α诱导活化的HaCaT细胞IL-6mRNA和IL-8mRNA的表达有下调作用,在药物浓度为10g/L和12.5g/L时,HaCaT细胞IL-10mRNA的表达量增强较为明显,与TNF-α诱导活化组相比,有显著性差异(P<0.05或P<0.01)。清热利湿饮对经TNF-α诱导活化的HaCaT细胞IL-8的分泌有不同程度的抑制作用,但对IL-10的影响不明显。结论:清热利湿饮可下调IL-6mRNA、IL-8mRNA的表达,上调IL-10mRNA的表达,且此作用随药物浓度的升高而增强;同时可降低细胞IL-8的分泌,但对IL-10的分泌无明显的作用。
OBJECTIVE: To investigate the mechanism of Qingrelihuyu decoction on the expression of IL-6mRNA, IL-8mRNA, IL-10mRNA and IL-8 and IL-10 in human immortalized keratinocytes HaCaT cells. Methods: HaCaT cells were used as research objects. The expression of IL-6mRNA, IL-8mRNA, IL-10mRNA and secretion of HaCaT cells induced by TNF-αwere observed by RT-PCR and ELISA respectively. IL-8, IL-10. Results: The Qingre Lishi Decoction could down-regulate the expression of IL-6mRNA and IL-8mRNA in HaCaT cells induced by TNF-α. At the concentration of 10g / L and 12.5g / L, the expression of IL-10mRNA Compared with TNF-α-induced activation group, there was a significant difference (P <0.05 or P <0.01). Qingreli wet drink inhibited the secretion of IL-8 in HaCaT cells induced by TNF-α to a certain extent, but had no obvious effect on IL-10. Conclusion: Qingre Lishi Decoction can down-regulate the expression of IL-6mRNA and IL-8mRNA, upregulate the expression of IL-10mRNA, and this effect increases with the increase of drug concentration; meanwhile, it can decrease the secretion of IL-8, -10 secretion no significant effect.
