目的:研究D101大孔吸附树脂分离纯化百蕊草总黄酮的工艺条件。方法:以总黄酮转移率和质量分数为指标,采用动态试验考察D101大孔吸附树脂对百蕊草总黄酮的分离纯化效果及影响因素。结果:D101大孔吸附树脂对百蕊草总黄酮的最佳纯化工艺条件为:上柱液总黄酮浓度为2.259 mg/mL,上柱液速度为1 BV/h,洗脱剂为70%乙醇,洗脱速度为1 BV/h,洗脱剂量为4 BV。总黄酮转移率为94.44%,质量分数为12.45%,此质量分数较百蕊草浸膏中总黄酮的含量(2.91%)提高了4倍多。结论:D101大孔吸附树脂对百蕊草总黄酮有显著的富集作用,且该纯化工艺简单稳定。
Objective: To study the conditions for the separation and purification of the total flavonoids of the microplants from D101 macroporous adsorption resin. Methods: Total flavonoid transfer rate and mass fraction were used as indices to study the separation and purification of total flavonoids from the D101 macroporous resin and its influencing factors by dynamic test. Results: The optimum conditions for the total flavonoids from the macroporous resin D101 for thesium were as follows: total flavonoid concentration of 2.259 mg / mL, column speed of 1 BV / h, eluent 70% ethanol , The elution rate of 1 BV / h, the elution dose of 4 BV. Total flavonoid transfer rate of 94.44%, mass fraction of 12.45%, this mass fraction than the total flavonoids in Thesberry extract (2.91%) increased more than 4 times. CONCLUSION: D101 macroporous resin has a significant enrichment effect on the total flavonoids of the micropyle and the purification process is simple and stable.
