目的 探讨CD2 8在系统性红斑狼疮 (SLE)患者外周血单个核细胞 (PBMC)中的表达水平及其意义。方法 应用反转录 聚合酶链反应 (RT PCR)检测了 34例活动期SLE患者和 30名正常人PBMC中CD2 8mRNA的表达水平。结果 活动期SLE患者CD2 8的阳性表达率为 2 0 6 % ,明显低于正常人对照组 (70 0 % ) ,差异非常显著 (P <0 0 0 1) ;活动期SLE组CD2 8的平均表达水平(0 19± 0 2 1)亦明显低于正常对照组 (0 43± 0 11) ,差异显著 (P <0 0 5 )。结论 CD2 8的异常表达可能在SLE发病机制中起作用 ,CD2 8mRNA的低水平表达可能与外周血CD2 8+ T细胞凋亡增加或迁移到炎症部位有关。
Objective To investigate the expression and significance of CD28 in peripheral blood mononuclear cells (PBMC) from patients with systemic lupus erythematosus (SLE). Methods The expression of CD28 in PBMC from 34 patients with active SLE and 30 normal controls was detected by reverse transcriptase-polymerase chain reaction (RT-PCR). Results The positive rate of CD28 expression in active SLE patients was 20.6%, which was significantly lower than that in normal controls (70.0%) (P <0.01) The expression level (0 19 ± 0 2 1) was also significantly lower than that of the normal control group (0 43 ± 0 11) (P <0.05). Conclusions The abnormal expression of CD28 may play a role in the pathogenesis of SLE. The low expression of CD28 may be related to the increase of apoptosis of CD8 + T cells or the migration to the inflammatory site.
