目的:建立一种以茜素紫为探针共振瑞利散射技术测定溶菌酶的方法。方法:唾液样品处理后,在优化测定条件下以茜素紫为探针共振瑞利散射技术测定其中的溶菌酶。结果:溶菌酶的检出限为0.43 mg/L(3S/N);溶菌酶标准溶液在2.0 mg/L~30.0 mg/L范围内线性良好,其相关系数分别为0.999;测定8.0 mg/L溶菌酶标准溶液的相对标准偏差(RSD)3.0%。样品加标回收率:97.2%~104.4%。结论:该方法灵敏度、准确度高;操作方便快速;具有较低的检出限,能满足测定人唾液中溶菌酶含量的检测工作。
OBJECTIVE: To establish a method for the determination of lysozyme using Alizarin violet as a probe by resonance Rayleigh scattering technique. Methods: Residual lysozyme was determined by resonance Rayleigh scattering technique with Alizarin violet as the probe after saliva sample treatment. Results: The detection limit of lysozyme was 0.43 mg / L (3S / N). The standard of lysozyme was linear in the range of 2.0 mg / L to 30.0 mg / L with the correlation coefficients of 0.999 and 8.0 mg / L The relative standard deviation (RSD) of lysozyme standard solution was 3.0%. Sample spike recovery: 97.2% ~ 104.4%. Conclusion: The method has high sensitivity and accuracy, convenient and fast operation, low detection limit, and can meet the detection of lysozyme in human saliva.
