观察15例老年呼吸系统感染患者体内奈替米星(Net)的药物动力学.用高效液相色谱-间接光度检测(HPLC-IPD)法,烟酰胺为检测剂,庚烷磺酸钠为反离子,测定单剂量和多剂量im及iv gtt 5mg/kg Net后的血清药物浓度,并计算主要药物动力学参数;同时测定了痰液药物浓度.结果:Net在体内消除半衰期为3.5h左右,单剂量im达峰时间0.77h,峰浓度为14.33mg/L.多剂量给药后体内轻微积蓄,痰药浓度与血药浓度无相关性.提示老年呼吸系统感染患者对Net消除较慢,生物利用度增高.
Pharmacokinetics of netilmicin (Net) in 15 elderly patients with respiratory infection was observed by HPLC-IPD method with nicotinamide as detector and sodium heptane sulfonate as anti Ion, the serum concentration of single and multiple doses of im and iv gtt 5mg / kg Net were measured, and the main pharmacokinetic parameters were calculated.Meanwhile, the drug concentration of sputum was also determined.Results: The half-life of Net in vivo was 3.5h, The single dose of im peak time 0.77h, the peak concentration of 14.33mg / L.
