目的观察祛风方对佐剂性关节炎大鼠的治疗作用及其机制。方法将32只Wistar大鼠随机分为正常组、模型组、激素组、中药组,每组8只。大鼠右后足趾皮内注射福氏完全佐剂0.05ml致炎;激素组造模后予强的松片每次0.25mg灌胃,每日2次;中药组造模后予祛风方每次3ml灌胃,每日2次。21天后检测各组大鼠膝关节滑膜白细胞介素1β(IL-1β)、白细胞介素6(IL-6)的mRNA表达水平。结果正常组、激素组、中药组大鼠膝关节滑膜组织IL-1β、IL-6 mRNA的表达均低于模型组,差异均有统计学意义(P<0.05);正常组、中药组与激素组3组之间比较无统计学差异(P>0.05)。结论佐剂性关节炎大鼠滑膜细胞IL-1β、IL-6 mRNA表达水平是上调的,激素和祛风方有抑制其转录和表达的作用。
Objective To observe the therapeutic effect of Qufeng Recipe on adjuvant arthritis in rats and its mechanism. Methods Thirty-two Wistar rats were randomly divided into normal group, model group, hormone group and TCM group, with 8 rats in each group. 0.05 ml of Freund’s adjuvant was infused intradermally in the right posterior toe of the rats to induce inflammation. After the model group of prednisone was given 0.25 mg intragastric administration twice daily, the Chinese medicine group was given Qufeng Decoction Each 3ml gavage, 2 times a day. Twenty-one days later, the mRNA expressions of interleukin-1β (IL-1β) and interleukin-6 (IL-6) in synovium of knee joints of rats in each group were detected. Results The expressions of IL-1β and IL-6 mRNA in synovium tissue of normal group, hormone group and traditional Chinese medicine group were lower than that of model group (P <0.05), while those in normal group, There was no significant difference between the three groups (P> 0.05). Conclusion The expression of IL-1β and IL-6 mRNA in synovial cells of rats with adjuvant arthritis is up-regulated, while the hormones and Qufeng Decoction can inhibit the transcription and expression of IL-1β and IL-6.
