目的:研究血虚肝郁证大鼠海马c GMP含量及其合成酶s GCα2、s GCβ1和水解酶PDE2A、PDE5A、PDE9A m RNA表达与芍药苷的干预作用。方法:分为空白组、模型组、40mg/kg芍药苷组、20mg/kg芍药苷组,造模第1天开始给药,空白组、模型组给等量蒸馏水,其余各组给予相应药物,共造模21d。治疗结束后,取脑组织,剥离海马,采用格里斯法(Griess法)测定NO含量、放射免疫法(RIA)测定c GMP含量以及实时荧光定量聚合酶链反应法(RT-q PCR)检测s GCα2、s GCβ1、PDE2A、PDE5A、PDE9A m RNA的表达。结果:血虚肝郁证模型大鼠海马的NO、c GMP含量升高且s GCα2、s GCβ1、PDE2A、PDE5A m RNA的表达增加(P<0.05,P<0.01),40mg/kg芍药苷能够降低NO、c GMP含量(P<0.01,P<0.001)且下调其合成与分解酶的表达。结论:芍药苷能够降低血虚肝郁证模型大鼠海马NO、c GMP的含量,且下调s GCα2、s GCβ1、PDE2A、PDE5A m RNA的表达,其机制与芍药苷影响合成酶与水解酶的变化有关,从两个方面综合作用保持其含量的动态平衡。
Objective: To study the effects of paeoniflorin on the content of c GMP and the expression of synthase s GCα2, s GCβ1 and PDE2A, PDE5A, PDE9A m RNA in hippocampus of rats with deficiency of blood and liver syndrome. Methods: The rats were divided into blank group, model group, 40 mg / kg paeoniflorin group and 20 mg / kg paeoniflorin group. The rats in the blank group and the model group were given the same amount of distilled water on the first day, A total of modeling 21d. At the end of treatment, the brain tissue was dissected, the hippocampus was dissected, the content of NO was determined by Griess method, the c GMP content by radioimmunoassay (RIA) and the real-time fluorescent quantitative polymerase chain reaction GCα2, s GCβ1, PDE2A, PDE5A, PDE9A m RNA expression. Results: The content of NO and c GMP in hippocampus of rats with blood deficiency and liver-qi stagnation increased and the expressions of s GCα2, s GCβ1, PDE2A and PDE5A m RNA increased (P <0.05, P <0.01), and 40 mg / kg paeoniflorin Reduce the contents of NO and c GMP (P <0.01, P <0.001), and down-regulate the expression of synthetic and degrading enzymes. CONCLUSION: Paeoniflorin can decrease the content of NO and c GMP in the hippocampus of rats with deficiency of blood and liver qi, and down-regulate the expressions of s GCα2, s GCβ1, PDE2A and PDE5A m RNA. The mechanism of this is that paeoniflorin affects the activity of synthase and hydrolase Change, from the two aspects to maintain the dynamic balance of its content.
