目的:研究从雷公藤中分离的喙枝孢属真菌Rhinocladiella sp.lgt-3的化学成分及其单胺氧化酶抑制活性。方法:采用各种色谱技术分离化合物,根据波谱数据鉴定化合物结构,采用酶标法测定化合物的单胺氧化酶抑制活性。结果:从Rhinocladiella sp.lgt-3中分离得到10个化合物,分别鉴定为:3,4-二氢-3,4,8-三羟基-1(2H)-四氢萘酮(1)、(3R,4R)-4-羟基蜂蜜曲菌素(2)、蜂蜜曲菌素甲醚(3)、4-(2-羟基丁炔氧基)苯甲酸(4)、2,4-二羟基-6-((R)-4-羟基-2-氧代戊基)-3-甲基苯甲醛(5)、(Z)-N-(4-羟基苯乙烯基)甲酰胺(6)、aspterric acid(7)、蟾毒色胺(8)、5-羟基-N,N-二甲基色胺-N-氧化物(9)、5-甲氧基-N,N-二甲基色胺(10)。化合物1、8~10显示了中等强度的单胺氧化酶抑制活性,其IC_(50)分别为32、36、50、56μmol/L。结论:其中,化合物3~10为首次从该属真菌中分离得到,化合物1的单胺氧化酶抑制活性为首次报道。
Objective: To study the chemical constituents and monoamine oxidase inhibitory activity of Rhinocladiella sp. Lgt-3 isolated from Tripterygium wilfordii. Methods: The compounds were isolated by various chromatographic techniques. Their structures were identified by spectroscopic data. The monoamine oxidase inhibitory activity of the compounds was determined by enzyme-linked immunosorbent assay. Results: Ten compounds were isolated from Rhinocladiella sp.lgt-3 and identified as 3,4-dihydro-3,4,8-trihydroxy-1 (2H) -tetralone (1) 3R, 4R) -4-hydroxy-aurin (2), honey aspartame (3), 4- (2- hydroxybutynyloxy) benzoic acid (4) (5), (Z) -N- (4-hydroxystyryl) carboxamide (6), aspterric acid (7), bufotin (8), 5-hydroxy-N, N-dimethyltryptamine-N-oxide (10). Compounds 1, 8-10 showed moderate intensity of monoamine oxidase inhibitory activity with IC 50 of 32, 36, 50, 56 μmol / L, respectively. CONCLUSION: Compounds 3 to 10 were isolated from this genus for the first time. The monoamine oxidase inhibitory activity of compound 1 was reported for the first time.
