目的:观察针刺百会对膜迷路积水模型大鼠耳蜗形态及水通道蛋白2(AQP2)表达的影响,并探讨其可能的调节机制。方法:将大鼠随机分为空白组、模型组、西药组及针刺组。除外空白组,其余各组大鼠采用腹腔注射血管加压素的方法复制膜迷路积水模型。空白组与模型组采取与针刺组相同的固定;西药组予托伐普坦溶液灌胃;针刺组予针刺百会治疗。各治疗组均每日治疗1次,连续治疗10d。干预结束后,行HE染色观察耳蜗积水程度,计算蜗管横截面积与蜗管加前庭阶横截面积之和的比值(R值);免疫组化检测耳蜗血管纹AQP2表达。结果:模型组R值较空白组显著增高(P<0.01);针刺组R值较模型组显著降低(P<0.01),与西药组比较,差异无统计学意义。模型组AQP2表达较空白组显著增强(P<0.01);针刺组AQP2表达较模型组显著减弱(P<0.05),与西药组比较,差异无统计学意义。结论:针刺百会可减轻大鼠膜迷路积水,其作用机制可能与下调耳蜗AQP2的表达有关。
Objective: To observe the effects of acupuncture at Baihui on the morphology of the cochlea and the expression of aquaporin 2 (AQP2) in rats with membranous labyrinth syndrome and to explore the possible regulatory mechanism. Methods: The rats were randomly divided into blank group, model group, western medicine group and acupuncture group. Except the blank group, the remaining rats in each group were injected with vasopressin by intraperitoneal injection of labyrinthine hydronephrosis model. The blank group and the model group took the same fixation as the acupuncture group; the western medicine group was treated with tolvaptan solution; the acupuncture group was given acupuncture Baihui treatment. Each treatment group were treated once daily for 10 days. After the intervention, HE staining was used to observe the degree of cochlear water retention, and the ratio of the cross-sectional area of the cochlea to the sum of the cross-sectional area of the cochlear and vestibule was calculated. The expression of AQP2 in the stria vascularis was detected by immunohistochemistry. Results: The value of R in the model group was significantly higher than that in the blank group (P <0.01). The value of R in the acupuncture group was significantly lower than that in the model group (P <0.01). There was no significant difference between the model group and the western medicine group. The expression of AQP2 in model group was significantly higher than that in blank group (P <0.01). The expression of AQP2 in acupuncture group was significantly weaker than that in model group (P <0.05). There was no significant difference between AQP2 and western medicine group. CONCLUSION: Acupuncture Baihui can reduce the membranous accumulation of water in rats, and its mechanism may be related to the down-regulation of AQP2 expression.
