著者已用细胞钓蛋白带技术证明,在备用根猫脊髓背角手术侧提取液电泳带相对迁移率(R;)0.11—0.12,045-0.48两处蛋白带中含有某些神经营养活性物质。为进一步验证该两蛋白带所含物质对神经元存活的影响,本文用改良的联合培养法观察手术Rfo.11-0.12、Rro.45—0.48两蛋白带对鸡胚背报节神经元存活的影响。具体方法如下:将常规的联合培养法(即将合某神经营养活性物质的凝胶小条放置于鸡胚DRG附近进行联合培养后,可见DRG的神经突起明显朝向凝胶小条的一面生长)改为将DRG的神经细胞悬液接种于涂有鼠尾胶原的24孔培养板孔内,并将磨碎的凝胶小条分别加入培养孔内。根据神经营养活性物质可从凝胶内折至培养液中的原理直接观察其对神经元存活的影响。结果显示:加入手术侧Rf0.11-0.12、Rr0.45-0.48蛋白带凝胶碎末的培养,其神经细胞存活率分别为082±0.05、081±0.02;而非手术侧组者其神经细胞存活率分别为0.55±0.07、0.54±0.08。分别比较两组的两带细胞存活率,均有显著性差异(P<0.01)。说明手术侧组Rf0.11-0.12及0.45—0.48两条蛋白带内均含有促进神经元存活的神经营养活性物质。
