目的建立反相高效液相色谱法同时测定人血浆中10种苯二氮类药物(氯氮、咪达唑仑、硝西泮、奥沙西泮、艾司唑仑、劳拉西泮、阿普唑仑、氯硝西泮、地西泮和三唑仑)。方法血浆样品经二氯甲烷萃取后,采用Shim-pack CLC-C8色谱柱,柱温35℃,流动相为0.01mol.L-1磷酸盐缓冲液-乙腈-无水乙醇-甲醇(68.2∶31.2∶0.3∶0.3),流速为1.10mL.min-1,紫外检测波长230nm。结果血浆中氯氮、咪达唑仑、硝西泮、奥沙西泮、艾司唑仑、劳拉西泮、阿普唑仑、氯硝西泮、三唑仑和地西泮在40~2000μg.L-1内线性关系良好(r≥0.9992),平均方法回收率均在97%~104%,萃取回收率均在70%以上,日内和日间RSD均≤11%(n=5)。结论本方法准确快速、简便易行,适用于人血浆中10种苯二氮类药物的同时检测。
OBJECTIVE To establish a method for the simultaneous determination of ten benzodiazepines in human plasma by reversed-phase high performance liquid chromatography (chlordiazepoxide, midazolam, nitrazepam, oxazepam, estazolam, lorazepam , Alprazolam, clonazepam, diazepam and triazolam). Methods The plasma samples were extracted with methylene chloride and analyzed by Shim-pack CLC-C8 column with a mobile phase of 0.01 mol·L-1 phosphate buffer-acetonitrile-absolute ethanol-methanol (68.2: 31.2 : 0.3: 0.3), flow rate of 1.10mL.min-1, UV detection wavelength of 230nm. Results Plasma levels of clonazepam, midazolam, nitrazepam, oxazepam, estazolam, lorazepam, alprazolam, clonazepam, triazolam and diazepam at 40 ~ 2000μg.L-1 (r≥0.9992), the average recoveries were both 97% -104% and the extraction recoveries were above 70% with RSDs less than or equal to 11% (n = 5) ). Conclusion The method is accurate, rapid, simple and easy to perform and is suitable for the simultaneous detection of 10 benzodiazepines in human plasma.
