目的了解外周血淋巴细胞微核标本制片的影响因素,寻找出最佳的室内质量控制制作条件。方法比较制片过程中不同的接种血量、培养温度、低渗时间以及离心速度对外周血中微核标本制作的影响,寻找最佳的室内质量控制制片效果。结果外周血微核制片过程中最佳的室内质量控制条件是接种血量是0.7×32规格注射器针头20~25滴;培养温度应该严格控制在(37.0±0.5)℃;最佳低渗时间是6~9min;最佳离心速度1500~2000 r/min;固定液应在使用前临时配置,放置1 h应废弃;玻片冷藏处理的效果比热水处理后的较好。结论外周血淋巴细胞微核标本制片的影响因素有接种血量、培养温度、低渗时间以及离心速度等,相关实验室予以关注,以确保其制备质量,提高微核检出准确率和读片效率,及时准确反映辐射损伤情况,对辐射防护作出正确的评价。
Objective To understand the influencing factors of peripheral blood lymphocyte micronucleus specimen preparation and to find the best conditions for the production of indoor quality control. Methods The effects of different inoculated blood volume, culture temperature, hypotonic time and centrifugation speed on the production of micronucleus specimens in peripheral blood were compared in the process of preparation to find out the best indoor quality control preparation. Results The optimal quality control conditions for peripheral micro-nucleus preparation were that the volume of inoculated blood was 20-25 drops of 0.7 × 32 syringe needle and the culture temperature should be controlled at (37.0 ± 0.5) ℃. The best hypotonic time Is 6 ~ 9min; the best centrifugal speed 1500 ~ 2000 r / min; fixed liquid should be temporarily configured before use, placed 1 h should be discarded; slide cold storage effect is better than after hot water treatment. Conclusion The influencing factors of peripheral blood lymphocyte micronucleus specimen production are vaccination blood volume, culture temperature, low osmotic time and centrifuge speed, etc. The relevant laboratories pay attention to ensure the preparation quality and improve the accuracy of micronucleus detection and read Film efficiency, promptly and accurately reflect the radiation damage, the radiation protection to make the correct evaluation.
