多胺是一类含有两个或两个以上氨基或亚氨基的长链脂肪族化合物,它包括腐胺、精脒、精胺及其衍生物。多胺广泛分布于生物体内,在生长活跃的组织中浓度较高,例如恶性肿瘤患者的瘤组织、血及尿中多胺含量增多。多胺对DNA、RNA及蛋白质的代谢都有重要影响。目前国外多用高效液相色谱法(HPLC)测多胺。国内曾有用柱前丹磺酰化反应分离多胺标准品的HPLC法报道。我们采用苯甲酰氯柱前衍生化的HPLC法测定生物样品中多胺则未见报道。本法反应条件简单,干扰物少,便于操作,并有较好的重复性和较高的灵敏度。
Polyamines are a class of long chain aliphatic compounds containing two or more amino or imino groups and include putrescine, spermidine, spermine, and derivatives thereof. Polyamines are widely distributed in the body and have a high concentration in actively growing tissues, such as tumor tissues, blood and urine polyamines in patients with malignant tumors. Polyamines have a major impact on DNA, RNA and protein metabolism. At present, foreign countries use multi-purpose high performance liquid chromatography (HPLC) to measure polyamine. China has used pre-column dansyl reaction polyamine standard HPLC method reported. We used benzoyl chloride precolumn derivatization HPLC determination of biological samples of polyamines have not been reported. The reaction conditions are simple, less interference, easy to operate, and has good repeatability and high sensitivity.
