目的探讨NF-κB信号通路蛋白在木脂素对胃癌SGC-7901细胞抗增殖作用中的机制。方法利用MTT、流式细胞术和Hochest染色、western blot方法检测木脂素对胃癌SGC-7901细胞增殖、凋亡、细胞周期和NF-κB信号通路相关蛋白p50和p65蛋白及bax和bcl-2蛋白表达的影响。结果 MTT结果表明,木脂可抑制胃癌SGC-7901细胞的增殖,并具有浓度和时间依赖性(P<0.05或P<0.01);流式细胞结果表明,木脂素促进细胞凋亡,与对照组相比有显著性差异(P<0.05),具有浓度依赖性,并阻止胃癌SGC-7901细胞从G0/G1期进入S期;western blot结果,木脂素下调p65、p50和bcl-2蛋白的表达而上调bax蛋白的表达。结论木脂素抑制胃癌SGC-7901细胞的增殖和凋亡与NF-κB信号通路密切相关。
Objective To investigate the mechanism of NF-κB signaling pathway in the anti-proliferation of gastric cancer SGC-7901 cells by lignan. Methods MTT, flow cytometry, Hochest staining and western blot were used to detect the expression of p50 and p65 protein and bax and bcl-2 protein in the proliferation, apoptosis, cell cycle and NF-κB signaling pathway of gastric cancer cell SGC-7901 Effect of protein expression. Results MTT results showed that lignan could inhibit the proliferation of gastric cancer SGC-7901 cells in a concentration- and time-dependent manner (P <0.05 or P <0.01). Flow cytometry showed that lignan promoted the apoptosis of SGC-7901 cells compared with the control (P <0.05), and it inhibited the proliferation of SGC-7901 cells from G0 / G1 phase into S phase. Western blot results showed that lignan down-regulated the protein levels of p65, p50 and bcl-2 But upregulated the expression of bax protein. Conclusion Lignans inhibit the proliferation and apoptosis of gastric cancer cell line SGC-7901 and NF-κB signaling pathway.
