本实验室建立了一株抗日本血吸虫抗原的单克隆抗体(CA7)。利用单抗亲和层析方法分离纯化了CA7靶抗原,该抗原分子量为220kd,糖蛋白染色阳性,蛋白酶水解不影响其与CA7的结合,高碘酸氧化能使之丧失与CA7结合的能力。碱水解去除多肽上的O-糖链后,分子量由220kd降为68kd,与单抗结合的能力也随之消失,提纯了含靶抗原决定簇的O-糖链,经Bio-gelP2柱层析及Dot-凝集素印迹实验结果表明:含CA7靶抗原决定簇的O-糖链是一个三糖结构,可能为半乳糖,N-乙酰氨基葡萄糖和N-乙酰氨基半乳糖。
The laboratory established a monoclonal antibody against Japanese schistosome antigen (CA7). The antigen of CA7 was isolated and purified by monoclonal antibody affinity chromatography. The antigen has a molecular weight of 220kd and positive staining for glycoprotein. Protease hydrolysis does not affect the binding of CA7 to CA7. L-periodate oxidation can lose its ability to bind to CA7. Alkaline hydrolysis to remove the peptide O-sugar chain, the molecular weight decreased from 220kd to 68kd, the ability to bind with the monoclonal antibody will disappear, purification of the target antigen-containing O-sugar chain by Bio-gelP2 column chromatography And Dot-Lectin blotting experiments showed that the O-sugar chain containing CA7 target epitope is a trisaccharide structure, which may be galactose, N-acetylglucosamine and N-acetylgalactosamine.
