目的探索依达拉奉对脑缺血再灌注大鼠脑红蛋白(Ngb)和14-3-3γ蛋白的影响。方法采用大脑中动脉闭塞法建立大鼠脑缺血再灌注模型,将48只雄性SD大鼠随机分为假手术组、模型组和依达拉奉预处理组;采用酶联免疫吸附法检测血清Ngb和14-3-3γ水平,采用蛋白免疫印迹、免疫组化检测脑组织中Ngb和14-3-3γ表达。结果模型组大脑和血清的Ngb和14-3-3γ水平均高于假手术组(P<0.05),依达拉奉预处理组大脑和血清的Ngb和14-3-3γ水平均高于模型组(P<0.05)。结论依达拉奉能通过上调脑缺血再灌注大鼠血清和脑组织中Ngb和14-3-3γ水平发挥脑保护作用。
Objective To explore the effect of edaravone on neuroglobin (Ngb) and 14-3-3γ protein in cerebral ischemia-reperfusion rats. Methods A rat model of cerebral ischemia-reperfusion was established by middle cerebral artery occlusion (MCAO). 48 male Sprague-Dawley rats were randomly divided into sham operation group, model group and edaravone preconditioning group. Serum was detected by enzyme-linked immunosorbent assay Ngb and 14-3-3γ levels, Western blotting was used to detect Ngb and 14-3-3γ expression in brain tissue by immunohistochemistry. Results The levels of Ngb and 14-3-3γ in brain and serum in model group were significantly higher than those in sham operation group (P <0.05). The levels of Ngb and 14-3-3γ in brain and serum of edaravone pretreatment group were higher than those of model group Group (P <0.05). Conclusion Edaravone can protect cerebral ischemia-reperfusion rats by increasing the level of Ngb and 14-3-3γ in serum and brain.
