目的:研究阳离子交换树脂分离和富集莲子心中黄酮与生物碱类物质的工艺条件。方法:以Nef、黄酮Ⅳ的吸附量及洗脱率为指标,对阳离子交换树脂的静态吸附-解吸性能、动态吸附-洗脱性能进行了考察。结果:药液比为1∶6的莲子心提取液经D152阳离子交换树脂吸附后,先用5BV纯化水洗去杂质,然后用10BV 70%乙醇洗脱,得黄酮类物质;用7BV 0.02 g/mL NaCl-70%乙醇洗脱,得生物碱类物质,两类物质洗脱率均在80%以上,精制倍数可分别达27.08倍及7.86倍。结论:该工艺能同时分离和富集莲子心中黄酮和生物碱类物质。 OBJECTIVE: To study the conditions for the separation and enrichment of flavonoids and alkaloids from lotus seeds by cation exchange resin. Methods: The static adsorption-desorption performance and dynamic adsorption-elution performance of cation exchange resin were investigated with the adsorption amount and elution rate of Nef and flavone Ⅳ as indexes. Results: The lotus root extract with liquid to liquid ratio of 1: 6 was adsorbed on D152 cation exchange resin. The impurities were first washed with 5BV purified water and then eluted with 10BV 70% ethanol to obtain flavonoids. 7BV 0.02 g / mL NaCl-70% ethanol, get alkaloids, two types of substance elution rate of more than 80%, respectively, refined times up to 27.08 times and 7.86 times. Conclusion: The process can simultaneously separate and enrich the lotus seed heart flavonoids and alkaloids.
