目的:建立测定血浆中的盐酸噻氯吡啶浓度的反相高效液相色谱法。方法:采用C18柱,流动相为甲醇水浓氨水(780∶220∶9)。200μl血浆用乙腈沉淀蛋白后,上清液直接进样。结果:本方法在0.1mg·L-1~2.0mg·L-1范围内线性良好,最低检测浓度为0.06mg·L-1。批间和批内误差均小于10%。方法回收率平均值为100.18%。结论:此方法具有操作简便、省时、分离度较好等特点。已经应用于正常人多剂量口服盐酸噻氯吡啶片剂后的相对生物利用度和药动学研究。
Objective: To establish a reversed-phase high performance liquid chromatography (HPLC) method for the determination of ticlopidine hydrochloride in plasma. Methods: The C18 column was used. The mobile phase consisted of methanol-water-concentrated ammonia (780: 220: 9). After 200 μl of plasma precipitated the protein with acetonitrile, the supernatant was injected directly. Results: The method was linear in the range of 0.1 mg · L-1 ~ 2.0 mg · L-1 with the lowest detection concentration of 0.06 mg · L-1. Inter-batch and intra-batch errors are less than 10%. The average recovery rate was 100.18%. Conclusion: This method has the characteristics of easy operation, time saving and good resolution. The relative bioavailability and pharmacokinetic studies have been applied to oral multidose oral ticlopidine hydrochloride tablets.
