目的:探讨不同缺氧时间点对小鼠心脏成纤维细胞旁分泌TGF-β1、BMP-2蛋白和mRNA的影响。方法:将体外分离培养的第2代小鼠心脏成纤维细胞,分别缺氧0、12、24、48 h。ELISA法和RT-PCR法检测不同时间点心脏成纤维细胞旁分泌TGF-β1、BMP-2的蛋白和mRNA水平。结果:相较于缺氧0 h,缺氧12 h后TGF-β1的蛋白和mRNA表达呈上调趋势,但24 h、48 h后逐步下调;而BMP-2在缺氧12 h、24 h后,蛋白和mRNA表达均逐步上调,48 h后mRNA水平下调。与缺氧24 h相比,缺氧48 h后TGF-β1和BMP-2的蛋白以及mRNA表达均下调。结论:不同缺氧时间诱导的心脏成纤维细胞旁分泌TGF-β1和BMP-2的变化,可能参与了心肌纤维化的病理过程。
Objective: To investigate the effects of different hypoxia time points on paracrine TGF-β1 and BMP-2 protein and mRNA in mouse cardiac fibroblasts. Methods: The second-generation mouse cardiac fibroblasts cultured in vitro were hypoxia for 0, 12, 24 and 48 h respectively. ELISA and RT-PCR were used to detect the protein and mRNA levels of paracrine TGF-β1 and BMP-2 in cardiac fibroblasts at different time points. RESULTS: Compared with hypoxia for 0 h, the protein and mRNA expressions of TGF-β1 increased 12 h after hypoxia, but down-regulated gradually after 24 h and 48 h, while BMP-2 decreased after 12 h and 24 h , Protein and mRNA expression were gradually increased, mRNA level down 48 h. Compared with 24 h hypoxia, the protein and mRNA expressions of TGF-β1 and BMP-2 were down-regulated 48 h after hypoxia. Conclusion: The changes of paracrine TGF-β1 and BMP-2 in cardiac fibroblasts induced by different hypoxia time may be involved in the pathological process of myocardial fibrosis.
