目的:通过采用限制片段差异显示聚合酶链反应(RFDD-PCR)建立卵巢癌组织与正常卵巢组织基因差异表达谱,分析比较卵巢癌发病机制相关基因的表达差异。方法:采集上皮性卵巢癌手术患者的卵巢癌组织和正常卵巢组织,用RFDD-PCR技术平行检测,对各组织间有表达差异的基因进行生物信息学分析,筛选与卵巢癌发病机制相关的基因。结果:运用RFDD-PCR技术,构建了两种不同组织的基因表达谱,通过对卵巢癌组织及正常卵巢组织差异表达谱的分析中发现与肿瘤的发生、发展密切相关的原癌基因和抑癌基因、细胞周期相关基因、增殖和凋亡相关基因、血管生长因子、代谢酶等6类基因36种因子的表达差异。结论:利用RFDD-PCR差异显示技术能够建立卵巢癌组织和正常卵巢组织基因差异表达谱,是对肿瘤的发病机制和寻找治疗靶点研究的一种有效的手段。
OBJECTIVE: To establish differentially expressed genes profiles of ovarian cancer tissues and normal ovarian tissues by restriction fragment PCR (RFDD-PCR) and to analyze the differences in the expression of genes involved in the pathogenesis of ovarian cancer. Methods: The ovarian cancer tissues and normal ovarian tissues of patients with epithelial ovarian cancer were collected and tested by RFDD-PCR. The genes with different expression among the tissues were analyzed by bioinformatics method, and the genes related to the pathogenesis of ovarian cancer . Results: Gene expression profiles of two different tissues were constructed using RFDD-PCR technique. The differential expression profiling of ovarian cancer tissues and normal ovarian tissues was found to be closely related to the occurrence and development of oncogenes and tumor suppressor Gene, cell cycle related genes, proliferation and apoptosis related genes, angiogenic growth factor, metabolic enzymes and other six kinds of 36 gene expression differences. Conclusion: The differential expression profiling of ovarian cancer tissue and normal ovary tissue using RFDD-PCR differential display technique is an effective means to study the pathogenesis of tumor and to find the therapeutic targets.
