本实验选用体重220~280g的雄性Wistar大鼠20只。取其左侧跖肌组织块做恒冷箱连续切片。将相邻切片做不同的组织化学反应,包括肌球蛋白ATPase、肌糖原、肌脂类、琥珀酸脱氢酶、细胞色素氧化酶、乳酸脱氢酶、6-磷酸果糖激酶及非特异性酯酶。用显微分光光度计对肌纤维的各种组织化学反应进行了定量测定。结果发现,根据大鼠跖肌肌球蛋白ATPase反应(碱性预孵育,pH10.4及酸性预孵育,pH4.65)可将肌纤维分为Ⅰ型、ⅡA型和ⅡB型3种纤维。各型纤维的其他组织化学反应不尽相同。糖原、乳酸脱氢酶、6-磷酸果糖激酶在Ⅱ型纤维分布较多;脂类、琥珀酸脱氢酶、细胞色素氧化酶在Ⅰ型和ⅡA型反应较强、反应强度的顺序为ⅡA>Ⅰ>ⅡB。非特异性酯酶的强度顺序为Ⅰ>ⅡA>ⅡB。所得结果说明,大鼠跖肌不同类型肌纤维存在着明显的代谢差异。其中ⅡA型纤维既具有很高的氧化活性,又具有很高的糖酵解活性,在骨骼肌能量代谢方面可能具有重要意义,值得深入研究。
In this experiment, 20 male Wistar rats weighing 220 ~ 280g were used. Take the left plantar muscle mass block to do continuous cooling box serial sections. The adjacent sections are subjected to different histochemical reactions including myosin ATPase, myogenin, myosties, succinate dehydrogenase, cytochrome oxidase, lactate dehydrogenase, 6-phosphofructokinase and nonspecific ester Enzymes. A variety of histochemical reactions of myofibers were quantitatively determined with a micro-spectrophotometer. The results showed that myofibers can be divided into three types Ⅰ, ⅡA and ⅡB according to the plantar muscle myosin ATPase reaction (alkaline preincubation, pH10.4 and acidic preincubation, pH4.65). Different types of fiber chemical reaction of different organizations. Glycogen, lactate dehydrogenase and 6-phosphofructokinase are more distributed in type II fibers. Lipids, succinate dehydrogenase and cytochrome oxidase are more reactive in type I and type IIA, and the order of reaction intensity is IIA > Ⅰ> ⅡB. The intensity of non-specific esterase order I> IIA> IIB. The results show that different types of rat plantar muscle fibers exist significant metabolic differences. Type IIA fibers have high oxidative activity and high glycolysis activity, which may be of great significance in the energy metabolism of skeletal muscle and deserve further study.
