Small structural modifications of tumor-associated carbohydrate antigens have the potential to find new anti-tumor vaccines. Since glucosidase is endowed with endoglucosidase tolerance and non-self-antigenic properties, the modification of carbohydrate antigens with S-linked glycosidic linkages instead of O-linked glycosidic linkages may result in a decrease of the weight of the native o-linked carbohydrate antigen Stronger immunogenicity, and may stimulate the production of antibodies that recognize natural oligosaccharides. To this end, we successfully completed the synthesis of a novel S-linked analog of the tumor associated disaccharide antigen STn.
