目的:研究氢分子对比格犬盆腹腔粘连形成的影响,并探讨其可能机制。方法:构建比格犬盆腹腔粘连模型,将15只比格犬随机均分为3组,分别为模型对照组、氢处理组、空白对照组,采用Phillips粘连评分标准进行形态学粘连程度评分;采用HE染色对肠管、子宫、卵巢组织形态进行镜下病理评价;造模前及造模后第1、4、7、14天检测外周血白细胞数目;ELISA法检测血清中CRP、IL-6、TNF-α含量;羟胺法检测超氧化物歧化酶水平(SOD);硫代巴比妥酸(TBA)比色法测丙二醛浓度。结果:模型对照组Phillips粘连评分显著高于空白对照组(n P<0.05);氢处理组Phillips粘连评分显著低于模型对照组(n P<0.05);氢处理组在镜下肠管、子宫、卵巢表面浆膜层增厚及炎症浸润程度低于模型对照组;模型对照组在造模后第1天各项炎症指标水平(白细胞计数、CRP、IL-6、TNF-α)及丙二醛水平均高于空白对照组(n P<0.05);SOD水平低于空白对照组(n P<0.05);氢处理组在造模后第1天各项炎症指标及丙二醛水平均低于模型对照组(均n P<0.05);SOD水平高于模型对照组(n P<0.05)。n 结论:氢分子能够有效抑制比格犬盆腹腔粘连形成,其可能机制是中和氧自由基,减轻炎症反应。“,”Objective:To study the effects of hydrogen molecule on the formation of pelvic and abdominal adhesions in dogs, and to explore its possible mechanism.Methods:The Beagle pelvic and abdominal adhesion model was constructed. 15 Beagle dogs were randomly divided into 3 groups: model control group, hydrogen treatment group and blank control group. The Phillips adhesion score was used to evalute the morphological adhesion degree. The morphology of intestinal, uterus and ovarian tissues was evaluated by hematoxylin-eosin (HE) staining. The inflammation biomarkers and oxygen free radical index were detected before modeling and on the 1st, 4th, 7th and 14th day after modeling. C-reactive protein (CRP), tumor necrosis factors-α (TNF-α) and interleukin(IL)-6 content in serum were detected by enzyme-linked immunosorbent assay (ELISA) kit; hydroxylamine method was used to detect the serum superoxide dismutase (SOD); thiobarbituric acid (TBA) colorimetric method was used to measure malondialdehyde concentration.Results:The Phillips adhesion score of the model control group was significantly higher than that of the blank control group (n P<0.05). The Phillips adhesion score of the hydrogen treatment group was significantly lower than that of the model control group (n P<0.05). In the hydrogen treatment group, the thickening of serous layer on the surface of intestine, uterus and ovary and the degree of inflammatory infiltration were lower than those in the model control group.The inflammatory indexes (leukocyte count, CRP, IL-6, TNF-α) and malondialdehyde level in the model control group were higher than those in the blank control group on the first day after modeling, and the superoxide dismutase (SOD) level was lower than that in the blank control group (alln P<0.05); the inflammatory indexes and malondialdehyde level in the hy-drogen treatment group were lower than those in the model control group on the first day after modeling (n P<0.05), and the SOD level was higher than that in the model control group (n P<0.05).n Conclusions:Hydrogen molecule can effectively inhibit the formation of pelvic adhesion in Beagle dogs, and its possible mechanism is to neutralize oxygen free radicals and reduce inflammatory reaction.
