目的:建立狗脊中多糖的含量测定方法,比较不同产地生制狗脊多糖的含量。方法:采用苯酚-硫酸法测定狗脊中多糖的含量,利用单糖和多糖的换算因子计算多糖含量,测定波长为489 nm。结果:在2.46~14.79 mg.L-1葡萄糖浓度C与吸光度A线性关系良好(r=0.999 5),平均回收率为98.7%,RSD 1.48%。不同产地的生狗脊多糖含量在2.48%~10.06%,制狗脊多糖含量在5.56%~15.17%。结论:该法简单、方便、准确、可靠,可以用来测定狗脊中多糖含量。不同产地狗脊中多糖的含量差异较大,其中福建产狗脊中多糖含量最高,狗脊经炮制后多糖的含量升高。
Objective: To establish a method for determination of polysaccharides in dog ridges, and to compare the content of polysaccharides in different regions of origin. Methods: The content of polysaccharide in dog ridge was determined by phenol - sulfuric acid method. The polysaccharide content was calculated by the conversion factor of monosaccharide and polysaccharide. The wavelength was 489 nm. Results: There was a good linear relationship between absorbance A and concentration C of glucose at 2.46-14.79 mg · L -1 (r = 0.999 5). The average recovery was 98.7% and the RSD was 1.48%. The content of polysaccharides in different areas was 2.48% ~ 10.06%, and the content of polysaccharides was 5.56% ~ 15.17%. Conclusion: The method is simple, convenient, accurate and reliable and can be used to determine the content of polysaccharides in dog ridges. The content of polysaccharides in dog ridges of different habitats varied greatly, among them, the content of polysaccharide in dog ridges in Fujian was the highest, and the content of polysaccharides in dog ridges increased after processing.
