目的:从升麻中分离得到升麻苷H-1,并以此为对照建立比色法测定升麻中升麻总皂苷含量方法。方法:以5%香草醛-高氯酸(1∶4)为显色体系,于60℃水浴中加热15min,置冰水中冷却2~3min,于537nm处测定吸光度。结果:样品在0.10mg~6.98mg(r=0.9995)范围内呈良好的线性关系,精密度、稳定性、重现性及加样回收率试验RSD均小于3%,平均加样回收率为100.27%。结论:该方法简便易行,稳定性、重现性好,可用于升麻中总皂苷的含量测定。
OBJECTIVE: To isolate and obtain the cimicifugin H-1 from cimicifuga and to establish a method for the determination of the content of cedil in cimicifuga by colorimetry. Methods: 5% vanillin perchloric acid (1: 4) was used as the color system and heated in a water bath at 60 ℃ for 15min, then cooled in ice water for 2 ~ 3min. The absorbance was measured at 537nm. The results showed that the RSDs of the samples were linear within the range of 0.10mg ~ 6.98mg (r = 0.9995). The RSDs of the precision, stability, reproducibility and recovery were less than 3% and the average recoveries were 100.27 %. Conclusion: The method is simple and easy to operate with good stability and reproducibility. It can be used for the determination of total saponins in Cimicifuga.
