目的观察不同浓度红景天苷对糖尿病大鼠血糖血脂及胰岛素的影响。方法采用小剂量链脲佐菌素加高脂高热量饲料喂养方法建立2型糖尿病大鼠模型,将建模成功后大鼠随机分为糖尿病模型(DM)组、罗格列酮组(1mg/kg)、红景天苷低剂量组(50mg/kg)、红景天苷中剂量组(100mg/kg)及红景天苷高剂量组(200mg/kg),另选10只为正常对照组(NS)。实验期间,每天记录大鼠摄食、饮水、精神状态;实验大鼠禁食12h,不禁水,大鼠尾静脉取血,血糖仪检测糖耐量,每2周一次。药物干预8周后,实验大鼠禁食12h,不禁水,麻醉后腹主动脉取血,检测空腹胰岛素。结果与糖尿病模型组比较,红景天苷治疗组大鼠空腹血糖及餐后2h血糖水平降低(P<0.05),胰岛素抵抗指数下降(P<0.05),总胆固醇水平降低(P<0.05)。结论红景天苷可降血糖血脂,改善胰岛素抵抗。
Objective To observe the effects of salidroside at different concentrations on blood glucose, blood lipid and insulin in diabetic rats. Methods A rat model of type 2 diabetes mellitus was established by using low-dose streptozotocin plus high-fat and high-calorie diet. The rats were randomly divided into diabetic model group (DM) and rosiglitazone group (1 mg / (50mg / kg), salidroside middle dose group (100mg / kg) and salidroside high dose group (200mg / kg), and the other 10 were normal control group (NS). During the experiment, rats were fed with food, water and mental state every day. The rats were fasted for 12h and were allowed to take water. The blood was taken from the tail vein of rats and the glucose tolerance was measured by blood glucose meter once every two weeks. After 8 weeks of drug intervention, the experimental rats were fasted for 12h, water was not allowed, and the abdominal aorta blood was taken after anesthesia to detect fasting insulin. Results Compared with diabetic model group, the fasting blood glucose and postprandial blood glucose level of salidroside treatment group decreased (P <0.05), insulin resistance index decreased (P <0.05) and total cholesterol level decreased (P <0.05). Conclusion Salidroside can reduce blood glucose, improve insulin resistance.
