试验以牛肺为原料提取肝素粗品,采用酶解结合氧化法精制肝素,提高肝素的效价,通过亚硝酸钠控制解聚法制备分子量在4000~7000D的低分子量肝素。通过正交试验考察了反应时间、亚硝酸钠浓度、溶液pH值等条件对解聚反应的影响。用红外图谱对其结构进行了鉴定,测定了产物的分子量,经Sephadex G-75凝胶色谱柱对解聚的低分子量肝素进行分离纯化。
The experiment used bovine lung as raw material to extract the crude heparin, purified the heparin by enzymolysis combined with oxidation method, increased the titer of heparin, prepared the low molecular weight heparin with molecular weight of 4000-7000D by sodium nitrite control depolymerization method. The effects of reaction conditions, sodium nitrite concentration and solution pH on the depolymerization reaction were investigated by orthogonal test. Its structure was identified by infrared spectrum. The molecular weight of the product was determined. The depolymerized low molecular weight heparin was separated and purified by Sephadex G-75 gel column.
