目的 了解北京地区 1990~ 1991年和 1997~ 1998年两个非连续的流行年中呼吸道合胞病毒(RSV)A、B亚型和基因型的流行情况。方法 用间接免疫荧光法 (IIF)检测RSV阳性鼻咽分泌物 (NPS)标本或RSV分离株 ,划分A、B亚型。根据N基因片段的限制性酶切图型将RSV分离株分成至少 6个基因型NP1- 6NP1,3和 6属于B亚型 ,NP2 4和 5属于A亚型。根据SH基因片段的核苷酸序列将A亚型分离株进一步划分为SH基因型SHL1- 6。SHL之间关系密切 ,并与NP有关。SHL1,3和 4紧密相关 ,属于NP2 ;SHL2和 6属于NP4;SHL5属于NP5。结果 1997冬季~1998年春季 145份RSVNPS标本中 ,83份 (5 7 2 % )为B亚型 ,6 2份 (4 2 8% )为A亚型 ,A∶B为 1∶1 3。1997~ 1998流行年所获的 10株RSV分离株中 ,2株为A亚型 ,8株为B亚型 ,A∶B为 1∶4。 1990~1991流行年所获的 10株毒株中 ,8株为A亚型 ,2株为B亚型 ,A∶B为 4∶1。1997~ 1998流行年分离到的 10株RSV分离株中 ,2株A亚型均属NP4,SHL2 ;而 2株B亚型均为NP3。根据NP推导出的A、B亚型划分与单克隆抗体定义的亚型一致。结论 北京地区RSVA、B亚型或多个基因型可同时流行 ,但每个流行年的优势基因型可不同。相距 5年仍可分离到相同的毒株 ,表明尽管A亚型毒株之间存在明显的差异性 ,一些基因型仍
Objective To understand the prevalence of subtype A and B of respiratory syncytial virus (RSV) in two non-consecutive epidemic years in Beijing from 1990 to 1991 and from 1997 to 1998. Methods RSV positive nasopharyngeal secretions (NPS) or RSV isolates were detected by indirect immunofluorescence (IIF), and subtype A and B were divided into two groups. RSV isolates were divided into at least 6 genotypes, NP1-6NP1, 3 and 6, according to the restriction map of the N gene fragment, to subtype B, and NP2 4 and 5 belong to subtype A, respectively. According to the nucleotide sequence of the SH gene fragment, the subtype A subtype was further divided into the SH genotype SHL1-6. SHL is closely related, and with the NP. SHL1, 3 and 4 are closely related and belong to NP2; SHL2 and 6 belong to NP4; SHL5 belong to NP5. Results Of the 145 specimens of RSVNPS from spring 1997 to spring 1998, 83 (57.2%) were subtype B and 62 (42.8%) were subtypes A and A: B were 1: 1. 3.1997 Among the 10 RSV isolates obtained in the 1998 ~ 1998 epidemic year, 2 were subtype A, 8 were subtype B, and A: B was 1: 4.
