目的:优选信前胡的大孔树脂纯化工艺条件。方法:以白花前胡乙素提取量为评价指标,通过正交试验考察乙醇体积分数、溶剂用量、提取时间对信前胡提取工艺的影响。通过单因素试验筛选大孔树脂型号并考察大孔树脂吸附与洗脱条件。采用HPLC测定白花前胡乙素含量,流动相甲醇-水(75∶25),检测波长321 nm。结果:最佳提取工艺为加8倍量60%乙醇提取3.5 h。选择X-5型大孔树脂,吸附时间60 min,白花前胡乙素吸附率92.36%,洗脱率71.24%,提取率98.71%,纯度8.88%。结论:X-5型大孔树脂适合分离纯化白花前胡乙素,为信前胡香豆素类成分的开发应用提供参考。
Objective: To optimize the process conditions for purifying macroporous resin from Xinqianhu. Methods: Taking the amount of pre-chlorhexidine extract as the evaluation index, the effects of ethanol volume fraction, solvent dosage and extraction time on the extraction process of Sinomenalis Obata were studied by orthogonal test. The single-factor experiments were used to screen the macroporous resin model and the conditions of macroporous resin adsorption and elution were investigated. HPLC method was used to determine the content of preanthemin. The mobile phase consisted of methanol-water (75:25) and the detection wavelength was 321 nm. Results: The best extraction process was adding 8 times the amount of 60% ethanol extraction 3.5 h. X-5 macroporous resin was selected and the adsorption time was 60 min. The adsorption rate of pre-chlorophyll a was 92.36%, the elution rate was 71.24%, the extraction rate was 98.71% and the purity was 8.88%. CONCLUSION: X-5 macroporous resin is suitable for the isolation and purification of pre-apocynin, providing a reference for the development and application of pre-apocynin composition.
