目的:观察通补奇经法中药复方对人子宫内膜基质细胞(Human endometrial stroma cells,hESCs)相关纤维连接蛋白(蛋白FN)、胰岛素样生长因子1(IGF-1)、催乳素(PRL)及纤溶酶原激活物抑制剂-1(PAI-1)表达的影响.方法:体外培养hESCs,采用MTT法发现安全中药复方浓度为10-4 mol/L,将实验分为溶剂对照组、甲孕酮(10-7 mol/L)组、甲孕酮(10-7 mol/L)+中药(10-4 mol/L)组、雌二醇(10-8mol/L)组、雌二醇(10-8 mol/L)+甲孕酮(10-7 mol/L)组、雌二醇(10-8 mol/L)+中药试剂(10-4mol/L)组、雌二醇(10-8 mol/L)+甲孕酮(10-7 mol/L)+中药(10-4 mol/L)组,采用ELISA方法检测各组子宫内膜基质细胞功能相关蛋白表达变化结果:与溶剂对照组比较,各组FN、IGF-1、PRL蛋白表达量均显著升高(P<0.05或P<0.01),而PAI-1表达显著减低,与甲孕酮组、雌二醇组分别比较,甲孕酮+中药组、雌二醇+甲孕酮组、雌二醇+中药组、雌二醇+甲孕酮+中药组FN、IGF-1、PRL蛋白表达量均显著升高(P<0.05或P<0.01),且甲孕酮+中药组、雌二醇+中药组、雌二醇+甲孕酮+中药组FN、IGF-1、PRL蛋白表达均分别高于对应的甲孕酮组、雌二醇组、雌二醇+甲孕酮组(P<0.05或P<0.01),而PAI-1呈现相反趋势.结论:通补奇经法中药复方可以促进人子宫内膜基质细胞相关蛋白的表达,其作用效果与甾体类激素相近.“,”Objective:To observe the effects of traditional Chinese medicine purgation-tonifying method compound prescription on expression of fibronectin(FN),insulin-like growth factor-1 (IGF-1),prolactin (PRL)and plasminogen activator inhibitor-1 (PAI-1)in human endometrial stroma cells (hESCs).Methods:hESCs were cultured in vitro.Determined by MTT method to find safe drug concentrations of 10-4 mol/L.Experiment can be divided into solvent control group,progestin (10-7 mol/L) group,progestin (10-7 mol/L) + Chinese medicine (10-4 mol/L) group,estradiol (10-8 mol/L),estradiol (10-8 mol/L) +progestin (10-7 mol/L)group,estradiol (10-8 mol/L)+ Chinese medicine (10-4 mol/L) group,estradiol (10-8 mol/L) +progestin (10-7 mol/L) + Chinese medicine (10-4 mol/L) group and solvent control group.Using ELISA method to detect each group of endometrial stromal cells function related protein expression changes.Results:Compared with solvent control group,each group FN,IGF-1,PRL,protein expression were significantly higher (P<0.05 or P<0.01),while PAI-1 expression significantly reduced potential.Compared with the armour of progesterone,estradiol group respectively,progesterone+Chinese medicine group,estradiol+progesterone group,estradiol+Chinese medicine group,estradiol+ progesterone + Chinese medicine group,FN,IGF-1,PRL protein expression were significantly higher (P<0.05 or P<0.01),while PAI-1 presents the opposite trend.Conclusion:Purgation-tonifying method compound prescription can promote the expression of basal cell related protein in human endometrium,and its effect is similar to that of steroid hormone.
