目的 探讨玻片法和试管法检测凝固酶的影响因素 .方法 按标准方法操作 ,比较两种血浆 (人血浆和兔血浆 )及三种抗凝剂 (EDTA Na2 ,EDTA K2 ,柠檬酸钠 )对结果的影响 .结果 使用柠檬酸钠和EDTA Na2 抗凝的兔血浆和人血浆 ,8株金黄色葡萄球菌均产生葡萄球菌凝固酶和聚集因子 ,而其它 2 6株葡萄球菌包括表皮葡萄球菌、溶血葡萄球菌、腐生葡萄球菌和人葡萄球菌不产生葡萄球菌凝固酶和聚集因子 .结论 柠檬酸钠和EDTA Na2 抗凝的兔血浆和人血浆检测葡萄球菌凝固酶均能得到正确结果 ,但EDTA K2 抗凝的人血浆不能用于检测游离凝固酶 .
Objective To investigate the influencing factors of coagulase by slide method and tube method.METHODS According to the standard method, two kinds of plasma (human plasma and rabbit plasma) and three kinds of anticoagulants (EDTA Na2, EDTA K2 and sodium citrate) were compared Results The effects of sodium citrate and EDTA Na2 anticoagulated rabbit plasma and human plasma, Staphylococcus aureus were all produced by Staphylococcus aureus and coagulase and aggregation factors, while the other 26 Staphylococcus aureus including Staphylococcus epidermidis, hemolysis Staphylococcus aureus, Staphylococcus saprophyticus and Staphylococcus aureus did not produce staphylococcal coagulase and coagulation factor.Conclusion Sodium citrate and EDTA Na2 anticoagulant rabbit plasma and human plasma staphylococcal coagulase enzyme can get the correct results, but EDTA K2 anti Coagulated human plasma can not be used to detect free coagulase.
