目的通过建立两次打击大鼠致多器官功能障碍综合征(MODS)的动物模型,探讨黄芪注射液对MODS致心肌和血管内皮细胞损伤的影响。方法将92只SD大鼠随机分为对照组、MODS组和治疗组。MODS组和治疗组复制大鼠MODS的动物模型,治疗组腹腔注入黄芪注射液,对照组和MODS组注入0.9%氯化钠溶液。分别在注射黄芪注射液和0.9%氯化钠溶液后24h,48h采集血液和心脏组织标本,测定大鼠血清中循环内皮细胞(CEC)数量、一氧化氮(NO)、内皮素-1(ET-1)、肌酸激酶同工酶(CK-MB)、肌酸激酶(CK)、乳酸脱氢酶(LDH)、心肌组织中肿瘤坏死因子-α(TNF-α)和细胞间黏附分子(ICAM-1)的水平,心脏组织标本行光镜下病理检查。结果治疗组大鼠腹腔注射黄芪注射液后24h和48hCEC数量、NO、ET-1水平明显低于相同时间点MODS组,差异有统计学意义(P<0.05);治疗组大鼠腹腔注射黄芪注射液后24h和48hTNF-α、ICAM-1、CK-MB、CK、LDH水平明显低于MODS组相同时间点水平,差异有统计学意义(P<0.05)。结论黄芪注射液对两次打击致MODS模型大鼠的心肌和血管内皮细胞损伤有明显的减轻和保护作用。
OBJECTIVE: To establish two animal models of multiple organ dysfunction syndrome (MODS) in rats and investigate the effect of astragalus injection on myocardial and vascular endothelial cell injury induced by MODS. Methods 92 SD rats were randomly divided into control group, MODS group and treatment group. MODS group and treatment group were duplicated the rat model of MODS. Astragalus injection was injected intraperitoneally into the treatment group, 0.9% sodium chloride solution was injected into the control group and the MODS group. Blood and heart tissue samples were taken at 24h and 48h after injection of astragalus injection and 0.9% sodium chloride solution, respectively. The levels of circulating endothelial cells (CEC), nitric oxide (NO) and endothelin-1 -1, CK-MB, CK, LDH, TNF-α and ICAM-1 in myocardium ICAM-1) levels, heart tissue specimens under light microscope pathological examination. Results At 24 and 48 hours after intraperitoneal injection of Astragalus injection, the levels of CEC, NO and ET-1 in the treatment group were significantly lower than those in the MODS group (P <0.05). The rats in the treatment group were injected intraperitoneally with astragalus injection The levels of TNF-α, ICAM-1, CK-MB, CK and LDH at 24h and 48h after operation were significantly lower than those at the same time point in MODS group, with statistical significance (P <0.05). Conclusion Astragalus injection significantly reduces and protects the myocardial and vascular endothelial cell injury of MODS model rats induced by two strikes.
