目的观察吡咯喹啉醌(PQQ)对许旺细胞神经生长因子(NGF)合成与分泌的影响,并对PQQ促损伤外周神经修复作用的可能机制予以探讨。方法将不同浓度PQQ(0.05mol/L、0.1mol/L、0.2mol/L)作用于原代培养、纯化的许旺细胞24h,应用ELISA与RT-PCR法分别于蛋白质和mRNA水平测定其对细胞NGF合成与分泌的影响。结果培养上清液中NGFA值分别为:对照组0.21±0.02;PQQ1组0.53±0.02;PQQ2组1.14±0.04;PQQ3组1.43±0.04;RT-PCRNGF产物A值/B-actinA值(PQQ1组1.36±0.04;PQQ2组1.71±0.01;PQQ3组2.24±0.03)均较对照组(0.83±0.02)高。不同浓度PQQ均可在蛋白质与mRNA水平显著促进许旺细胞合成与分泌NGF。结论PQQ可在mRNA水平及蛋白质水平促进许旺细胞合成与分泌NGF,这可能足其促损伤神经修复作用的机制之一。
Objective To investigate the effect of pyrroloquinoline quinone (PQQ) on the synthesis and secretion of nerve growth factor (NGF) in Schwann cells and to explore the possible mechanism of PQQ on promoting peripheral nerve repair. Methods Different concentrations of PQQ (0.05mol / L, 0.1mol / L, 0.2mol / L) were applied to primary cultured Schwann cells for 24 hours. ELISA and RT-PCR were used to determine the protein and mRNA levels Effect of Cell NGF Synthesis and Secretion. Results The NGFA values in the culture supernatants were 0.21 ± 0.02 in the control group, 0.53 ± 0.02 in the PQQ1 group, 1.14 ± 0.04 in the PQQ2 group, 1.43 ± 0.04 in the PQQ3 group, and 1.36 ± 0.04 in the PQQ1 group ± 0.04; PQQ2 group 1.71 ± 0.01; PQQ3 group 2.24 ± 0.03) were higher than the control group (0.83 ± 0.02). Different concentrations of PQQ can significantly promote the synthesis and secretion of NGF in Schwann cells at protein and mRNA levels. Conclusion PQQ can promote the synthesis and secretion of NGF in Schwann cells at the level of mRNA and protein, which may be one of the mechanisms that promote the repair of nerve.
