目的:比较左金方、黄连、吴茱萸对人胃癌细胞SGC-7901生长的活性抑制和凋亡诱导作用。方法:采用MTT比色法观察左金方、黄连、吴茱萸对SGC-7901的生长抑制情况;流式细胞仪PI单染亚二倍体峰分析细胞凋亡率;Hoechst 33258染色观察细胞形态学变化以及Western blot方法检测左金方、黄连、吴茱萸对人胃癌细胞SGC-7901中Bcl-2、Bax蛋白表达的影响。结果:左金方和黄连分别作用SGC-7901细胞后,均能抑制细胞活性,且与时间呈依赖关系;1.0 mg·m L-1左金方和对应浓度的黄连(0.857 mg·m L-1)作用于SGC-7901细胞48 h,细胞凋亡率分别为(36.40±1.59)%和(21.90±0.27)%,与对照组(1.69±1.91)%比较差异均有统计学意义(P<0.01),组间比较差异均有统计学意义(P<0.01),对应浓度的吴茱萸(0.143mg·m L-1)几无作用;Hochest染色细胞出现核固缩状和颗粒状荧光等典型的凋亡学特征;左金方和黄连均可降低SGC-7901细胞Bcl-2基因表达和增加Bax基因表达。结论:左金方与黄连均可通过上调Bax/Bcl-2的比率诱导胃癌细胞的凋亡,但左金方的作用优于黄连(P<0.01),吴茱萸几无作用。
OBJECTIVE: To compare the inhibitory effect of Zuojinfang, Coptis chinensis and Evodia on the growth inhibition and apoptosis induction of human gastric cancer cell SGC-7901. Methods: MTT colorimetry was used to observe the growth inhibition of SGC-7901 by Zujinfang, Coptis chinensis and Evodia rutaecarpa. Flow cytometry was used to analyze the apoptosis rate of the cells by sub-diploid peak of PI single staining. Hoechst 33258 staining was used to observe the morphological changes Western blot was used to detect the effects of Zujinfang, Coptis chinensis and Evodia on the expression of Bcl-2 and Bax in human gastric cancer cell line SGC-7901. RESULTS: After treated with ZJF and Coptis respectively, the activity of SGC-7901 cells was inhibited and the activity of SGC-7901 cells was dependent on the time. The effects of ZQF on SGC-7901 cells and the corresponding concentrations of Coptis chinensis (0.857 mg · m L- 1) in SGC-7901 cells for 48 h, the apoptotic rates were (36.40 ± 1.59)% and (21.90 ± 0.27)%, respectively, which were significantly different from those in control group (1.69 ± 1.91)% 0.01). There was significant difference between the two groups (P <0.01), but there was no effect in the corresponding concentration of Evodia rutaecarpa (0.143 mg · m L-1). The typical Hochest cells showed nuclear pyknosis and granular fluorescence Apoptosis characteristics; Zuojinfang and Coptis can both reduce Bcl-2 gene expression and increase Bax gene expression in SGC-7901 cells. Conclusion: Zuojinfang and Coptis can induce the apoptosis of gastric cancer cells by up-regulating the ratio of Bax / Bcl-2, but Zujinfang is better than Coptis (P <0.01) and Evodia has no effect.
