目的:对蝎毒抗癌多肽对肝癌细胞株中癌症高表达蛋白Hec1抑制作用进行研究。方法:小鼠肝癌细胞株Hec1和人肝癌细胞株SMMC-7721。阳性对照选用5-Fu(10μg/ml),蝎毒抗癌多肽选用3个浓度(0.09μg/ml、0.06μg/ml和0.03μg/ml)。结果:蝎毒抗癌多肽的IC50剂量效应关系不明显,细胞毒性最强。随着蝎毒抗癌多肽作用时间的延长,其对肝癌细胞株中癌症高表达蛋白Hec1抑制作用也日益增大。结论:蝎毒能够对肝癌细胞株中的游离钙水平进行影响,能够降低细胞分裂信使的钙离子水平,还能够有效抑制住肝癌细胞株中癌症高表达蛋白Hec1增殖。
Objective: To study the inhibitory effect of anticancer polypeptide from scorpion venom on Hec1, a highly expressed protein in cancer cell lines. Methods: Mouse hepatoma cell line Hec1 and human hepatocellular carcinoma cell line SMMC-7721. Positive control selected 5-Fu (10μg / ml), scorpion venom anticancer peptides selected three concentrations (0.09μg / ml, 0.06μg / ml and 0.03μg / ml). Results: The IC50 of scorpion venom anticancer peptide dose-effect relationship is not obvious, the strongest cytotoxicity. As the scorpion venom anti-cancer peptide effect of time, its hepatocellular carcinoma cell line highly expressed protein Hec1 inhibitory effect is also increasing. Conclusion: Scorpion venom can affect the level of free calcium in hepatocellular carcinoma cell lines, reduce the level of calcium in cell division messenger, and inhibit the proliferation of Hec1, a highly expressed cancer cell line.
