目的:建立同时测定山西、甘肃等不同省区产远志中远志酮Ⅲ、3,6’-二芥子酰基蔗糖、细叶远志皂苷A 3种成分含量的HPLC测定方法。方法:超声提取,ODS-C18色谱柱,柱温35℃,流动相为0.05%磷酸乙腈溶液(A)-0.05%磷酸水溶液(B),梯度洗脱;检测波长315 nm。结果:远志中3种成分在30 min内良好分离,其它成分在65 min内基本分离。3种成分在0.002~1.0μg质量范围内线性关系良好、平均加样回收率95.3%~109.0%。结论:不同产地中远志酮Ⅲ、3,6’-二芥子酰基蔗糖、细叶远志皂苷A含量范围分别为1.01~7.47、2.44~5.63、2.13~7.19 mg·g~(-1)。方法操作简便,结果准确,可用于远志药材3种成分含量测定。
OBJECTIVE: To establish a HPLC method for the simultaneous determination of three constituents of Polygalaectonone Ⅲ, 3,6’-acesulfonyl Sucrose, Prunella sylvestris L. from different provinces in Shanxi, Gansu and other provinces. Methods: ODS-C18 column was used for ultrasonic extraction. The column temperature was 35 ℃. The mobile phase consisted of 0.05% phosphoric acid solution (A) and 0.05% phosphoric acid solution (B). The detection wavelength was set at 315 nm. Results: The three constituents of Polygala tenuifolia were well separated within 30 min, while the other constituents were separated within 65 min. The linearity of the three components in the range of 0.002 ~ 1.0μg was good with average recoveries of 95.3% -109.0%. CONCLUSION: The contents of Ⅲ, 3,6’-aescinylsucrose and Anemarrhena acutissima A from different areas were 1.01 ~ 7.47, 2.44 ~ 5.63 and 2.13 ~ 7.19 mg · g -1, respectively. The method is simple, accurate and can be used to determine the content of three components of Polygala tenuifolia.
