以蚜虫传染发生云南烟草丛枝症病害的烟株为材料进行组织培养,连续培育3代,以第3代组培烟苗为毒源经蚜虫传染健康烟株,提取毒源和各实验烟株的总核酸进行琼脂糖凝胶电泳.结果表明:第3代组培烟苗为毒源经蚜虫传染的烟株发病表现丛枝症,并检测到与云南烟草丛枝症病害相关的低分子量RNA.说明与云南烟草丛枝症病害相关的病原能在寄主离体培养组织中长期保存,成功建立云南烟草丛枝症病害的标准毒株.
Aphid-infected tobacco plants with tobacco witches-bush disease in Yunnan were used as materials for tissue culture. Three generations of tobacco plants were continuously cultured. The third generation of tobacco seedlings were used as poison sources to transmit healthy tobacco plants through aphids. The total nucleic acid was subjected to agarose gel electrophoresis. The results showed that the tobacco plants infected by aphids in the 3rd generation of tobacco seedlings were witches’ sticks, and the low molecular weight RNAs associated with the disease of Yunnan bush were detected. The results indicated that the pathogen related to the disease of Yunnan bush could be stored for long time in vitro and the standard strain of Yunnan bush disease was successfully established.
