目的研究竹红菌甲素(hypocrellin A,HA)诱导人黑色素瘤(A375-s2)细胞死亡的分子生物学机制。方法应用结晶紫染色法、流式细胞技术及RT-PCR技术等。结果HA能够诱导A375-S2细胞产生凋亡,并诱导细胞周期停滞在S期,同时诱导细胞周期蛋白CyclinB1的mRNA表达增加。结论HA诱导细胞周期因子CylinB1 mRNA表达增加与细胞周期停滞在S-期密切相关,同时细胞周期的阻碍可能是诱导A375-S2细胞凋亡的一个重要原因。
Objective To investigate the molecular mechanism of hypocrellin A (HA) induced human melanoma (A375-s2) cell death. Methods using crystal violet staining, flow cytometry and RT-PCR technology. Results HA could induce apoptosis in A375-S2 cells and induce cell cycle arrest in S phase while inducing an increase in the expression of cyclinB1 mRNA. Conclusion The increased expression of CylinB1 mRNA induced by HA is closely related to the cell cycle arrest in S phase, and the cell cycle arrest may be an important reason for inducing A375-S2 cell apoptosis.
