目的研究血管内皮生长因子(vascularendothelialgrowthfactor,VEGF)对人胚胎视网膜血管发生的调节作用。方法收集54例9~40周龄胎儿眼球后壁标本,免疫组织化学染色,光镜观察。结果①VEGF在视网膜的表达呈波峰式分布,高峰在9~13周及26周左右。②节细胞层的梭形细胞(血管内皮细胞前体细胞)、血管内皮细胞呈增殖细胞核抗原(proliferationcelnuclearantigen,PCNA)免疫反应阳性,水平波动,高峰在9~13周及21周前后,此期间梭形细胞不断增殖、分化形成内皮细胞索,经改建形成视网膜内层血管,26、34周起见内核层内、外缘血管内皮细胞呈PCNA免疫反应阳性,并保持至足月。③视网膜VEGF表达量与梭形细胞、血管内皮细胞PCNA表达量呈显著正相关(r=0.736,P<0.01)。结论VEGF对视网膜血管的发生有显著促进作用
Objective To investigate the regulatory effect of vascular endothelial growth factor (VEGF) on human embryonic retinal vascularization. Methods 54 cases of fetus from 9 to 40 weeks fetuses were retrospectively studied by immunohistochemical staining and light microscopy. Results ① The expression of VEGF in the retina showed a peaked distribution with peaks at 9-13 and 26 weeks. (2) The spindle cells (vascular endothelial progenitor cells) and vascular endothelial cells (PCNA) were positive for immunoreactive cells in the cell layer, the level fluctuated between 9-13 weeks and 21 weeks before and after this period Cells continue to proliferate and differentiate to form endothelial cell cords, which are reconstructed to form inner retinal vessels. At week 26 and week 34, the endothelial and peripheral vascular endothelial cells showed positive PCNA immunoreactivity and remained full term. The expression of VEGF in retina was positively correlated with the expression of PCNA in spindle cells and vascular endothelial cells (r = 0.736, P <0.01). Conclusion VEGF can significantly promote the occurrence of retinal blood vessels
